Molecular identification and phylogenetic positions of medicinal Plantago species in the genus Plantago based on universal DNA barcoding

초록

The identification of accurate plant species is vital role for the quality control and standardization of herbal medicines. Due to the morphological similarities of medicinal parts, it is very difficult to distinguish authentic medicinal materials from other closely related species. The herbal medicine Plantaginis Semen, Plantato Seed, is described to include only the ripe seeds of Plantago asiatica L. or Plantago depressa Willd. in the Korean pharmacopoeia. However, the accurate identification of these authentic species from other Plantago species is very difficult using morphological characteristics of seeds. To establish a reliable discrimination tool distinguishing authentic species from adulterants, we analyzed universal DNA barcodes, nrDNA-ITS, matK, psbA-trnH, and trnL-F, and obtained species-specific nucleotides enough to identify authentic Plantago species from adulterants. To verify the phylogenetic relationships between authentic and inauthentic species in the genus Plantago, furthermore, we constructed phylogenetic tree using DNA barcode sequences of six Plantago species distributing in Korea. However, the phylogenetic analyses revealed that the six species did not distinctly classified corresponding to the species levels and the authentic Plantago species did not showed significant phylogenetic correlation among the species.
