Genotyping-by-sequencing (GBS) reveals population genetic diversity and differentiation of narrow Korean endemic Phedimus latiovalifolium (Crassulaceae)

초록

Phedimus latiovlaifolium has been reported to occur sporadically in high mountain areas of Gangwon-do Province, since it was first described by YN Lee (1992) from Geumdae-bong Peak area of Taebaek Mountain. Morphologically, it resembles P. ellacombianus, but can be distinguished from the latter by broadly ovate or rhombic leaves forming terminal rosettes. Although P. latiovalifolium was originally suggested as potential hybrid origin between two sympatric species P. aizoon and P. kamtschaticus, the hybrid origin hypothesis was refuted based on lack of allozyme additive profiles from two putative parental species as well as morphological divergence. In addition, our recent chloroplast phylogenomic analysis suggested that P. latiovalifolium is a distinct taxon from congeneric species in Korea. In this study, we estimated population genetic diversity and differentiation of four populations (Dosol Mountain, Daegwanryong Pass, Manhang-jae Pass, and Geumdae-bong Peak) of P. latiovalifolium in comparison with representative populations of congeneric species (3 populations of P. kamtschaticus, 2 populations of P. aizoon, and 2 populations of P. ellacombianus) based on GBS data. A total of 18,170 SNPs showed that P. latiovalifolium is genetically differentiated from its congeneric species based on PCoA, UPGMA tree and STRUCTURE analysis. However, genetic differentiation was not evident among four populations without any correlation between genetic and geographic distances. This study corroborates the taxonomic distinction of P. latiovalifolium and provides degree of population genetic diversity and differentiation for management and conservation of highly vulnerable high mountain species to climatic change.
